Materials Needed

Protocol

  1. Add sterile water to final volume of 250μL.
  2. Add 250μL of phenol:chloroform:isoamyl alcohol.
  3. Invert & centrifuge 2 min at 15 000 rpm.
  4. (Keep the aqueous/top layer!) Pipet the aqueous layer into a new tube.
  5. Add 250μL chloroform.
  6. Invert. Spin 1 min at 15 000 rpm.
  7. Pipet the aqueous layer into a new tube.
  • You can precipitate this prep to concentrate the DNA if desired.
