Materials Needed

Protocol

6.25 mM NH4NO3, 1.84 mM KH2PO4, 3.35 mM KCl, and 1.01 mM MgSO4, adjust pH to 7.5 and autoclave for 20 min at 121C.[1]

After autoclaving, add sterile glucose to a final concentration of 2.77 mM and sterile adult bovine serum to a final concentration of 30% (v/v).[2]

Serum SAPI-medium is stable up to a year when stored at -20C. Do not refreeze after thawing.
