supE44 (synonymous to glnV) acts as an amber nonsense mutation (insertion of UAG stop codon) suppressor. This tRNA structural gene mutation, first isolated by Bachmann et al., 1976, causes the transition of the GUC glutamine anticodon to a AUC anticodon. The mutant SupE44 tRNA competes with the amber nonsense mutation tRNA in translation, yielding a phenotype with lower levels of termination of translation.
supE44 's phenotype was determined by Inokuchi et al., 1979. Upon infection with mutant phage containing supE44, higher levels of tRNAGln2 were noted; nuclease digestion and gel analysis yielded similar results to Yaniv and Folk's (1975) tRNAGln2 sequences.
